DNA sequencing and DNA profiling

Cross-contamination of cell lines is frequent in cell cultures. Moreover, genomic instability and chromosomal abnormalities can occur after extended passaging of cell lines. Several studies show that cells after multiple passages develop genetic mutations, including duplications and deletions. As a consequence, cell banks and cell factories must control cell line genomic stability and monitor cell line identity to avoid cell contamination. To this aim, Procelltech has developed molecular methods based on DNA fingerprinting to detect cell line contamination and on DNA sequencing to verify genomic instability. These assays include microsatellite or short tandem repeat (STR) analysis, single nucleotide polymorphism (SNP) analysis and DNA sequencing. These molecular assays are a rapid, reproducible and high sensitivity and are conducted in the EU (EP 6.0., 2.6.21).
